In my opinion, this really comes down to personal preference. The XRC model feels more like the traditional Windows GUI model, where the dialog layouts are hidden away in a separate resource file.
For me, I tend to use XRC to experiment with my layouts. When I get a layout that I like, I go write out the equivalent wxPython code by hand. That's partly superstition, partly a feeling of control, and partly my fondness for "one file solutions". There are no compelling technical reasons for my choice.
